Susanne Herold, PhD<\/h3>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"alignleft wp-image-20120 size-full\" src=\"https:\/\/ilh-giessen.de\/wp-content\/uploads\/2021\/04\/Herold_1.jpg\" alt=\"\" width=\"311\" height=\"162\" \/><\/p>\n<p>Susanne Herold is W3 Professor for Internal Medicine, Infectious Diseases and Pulmonary Research and Director of the Department of Medicine V, UKGM in Giessen. She obtained her medical and PhD degree at Justus Liebig University of Giessen, Germany. Her research focuses on macrophage-epithelial interactions during viral pneumonia induced by influenza virus and SARS-CoV-2. She addresses mechanisms of tissue injury and regeneration processes in this context and aims at translating these findings into clinical applications. She runs several multi-center investigator-initiated trials aiming at clinical improvement of patients with viral pneumonia and ARDS by targeted interventions. She serves on the Board of Directors of the Excellence Cluster EXC2026 Cardiopulmonary Institute (CPI), and coordinates the disease area \u201cPneumonia and Acute Lung Injury\u201d at the German Center for Lung Research (DZL).<\/p>\n<p>For more information visit: <a href=\"https:\/\/www.pulmonary-infections.com\/\" target=\"_blank\" rel=\"noopener\">https:\/\/www.pulmonary-infections.com\/<\/a><\/p>\n<p>The Herold lab provides high throughput cell separation and analysis tools to address cell-cell interaction networks at high resolution up to single cell level in acute, chronic, and malignant lung diseases, with a focus on macrophage reprogramming into functional phenotypes and their interaction with parenchymal lung cells impacting disease outcome. Technologies include FACS analysis (BD FACSCanto<sup>TM<\/sup>, BD LSRFortessa<sup>TM<\/sup>) and MACS\/FACS-based cell\u00a0separation (Miltenyi autoMACS\u00ae Separator; BD cell sorter: FACSAria<sup>TM<\/sup> III, BD FACSMelody<sup>TM<\/sup>, BD FACSDiscover<sup>TM<\/sup> S8), allowing\u00a0bulk, single cell, plate, as well as index sorting for downstream applications such as sc\/snRNA sequencing techniques (10x Genomics Chromium and Bio-Rad Droplet Digital<sup>TM<\/sup> Sequencing (ddSEQ<sup>TM<\/sup>) platforms), cell culture including organoid cultures, for analysis of cells from in vitro\/ex vivo cultures, from circulating cell pools, and from complex healthy and diseased murine\/human lung tissues. Using these technologies, we will address the following topics within the consortium in a collaborative effor<\/p>\n<p>&nbsp;<\/p>\n<p><img loading=\"lazy\" decoding=\"async\" class=\"wp-image-22910 alignnone\" src=\"https:\/\/ilh-giessen.de\/wp-content\/uploads\/2024\/11\/2024_SH-300x230.jpg\" alt=\"\" width=\"1323\" height=\"1014\" srcset=\"https:\/\/ilh-giessen.de\/wp-content\/uploads\/2024\/11\/2024_SH-300x230.jpg 300w, https:\/\/ilh-giessen.de\/wp-content\/uploads\/2024\/11\/2024_SH.jpg 593w\" sizes=\"auto, (max-width: 1323px) 100vw, 1323px\" \/><\/p>\n<p><strong>Service<\/strong><\/p>\n<p>Our FACS Facility is located at our labspace at the Biomedizinisches Forschungszentrum Seltersberg (BFS) where we offer collaborations using state-of-the-art flow cytometry equipment and assistance with respect to experimental design regarding user specific analysis and sorting needs, including sample preparation, acquisition and data analysis.<\/p>\n<p><strong>Instruments<\/strong><\/p>\n<p><em><strong>BD FACSDiscover<sup>TM<\/sup> S8 Cell Sorter<\/strong><\/em><\/p>\n<p>Our recently acquired FACSDiscover<sup>TM<\/sup> S8 is an advanced spectral cell sorter equipped with 5 lasers (355 nm, 405 nm, 488 nm, 561 nm, and 640 nm) capable of measuring up to 30 fluorescent parameters. The system features multiple nozzle options (85 \u00b5m, 100 \u00b5m, and 130 \u00b5m) to handle a range of cell sizes. It allows for sorting of up to six populations (85 \u00b5m nozzle) simultaneously or plate sorting and supports single-cell index sorting. The FACSDiscover<sup>TM<\/sup> S8 is the only cell sorter offering image-enabled spectral sorting using BD SpectralFX<sup>TM<\/sup> technology. The SpectralFX<sup>TM<\/sup> technology allows visual confirmation on cell populations in real time during analysis and sorting. Therefore, this machine is ideal for high-parameter flow cytometry applications, including detailed phenotyping, functional assays, and single-cell RNA sequencing sample preparation.<\/p>\n<p><strong><em>BD FACSMelody Cell Sorter<\/em><\/strong><\/p>\n<p>Our FACSMelody<sup>TM<\/sup> is a benchtop flow cytometer designed for easy and precise cell sorting. It is equipped with three lasers (488 nm, 640 nm, and 405 nm) and can measure up to 9 fluorescent parameters. The system features the 100 \u00b5m nozzle which is suitable for most cell types. It is capable of performing bulk (4-way sorting) as well as plate sorting with single-cell index sorting. The BD FACSMelody<sup>TM<\/sup> is also suitable for applications such as single-cell RNA sequencing sample preparation, offering reliable performance and user-friendly operation to streamline your research workflow.<\/p>\n<p><em><strong>BD FACSAria<sup>TM<\/sup> III Cell Sorter<\/strong><\/em><\/p>\n<p>Our FACSAria<sup>TM<\/sup> is an advanced cell sorter equipped with four lasers (488, 561, 633, 405 nm) capable to measure up to 16 fluorescent parameters. Four different nozzles (70, 85, 100 and 130 \u00b5m) can be used depending on cell size. The system features bulk (4-way sorting) as well as plate sorting with single-cell index sorting. This machine is also ideal for single-cell RNA sequencing sample preparation.<\/p>\n<p><strong><em>BD LSRFortessa<sup>TM<\/sup> and FACSCanto<sup>TM<\/sup> Analyzers<\/em><\/strong><\/p>\n<p>Our LSRFortessa<sup>TM<\/sup> and FACSCanto<sup>TM<\/sup> flow cytometry analyzer are equipped with four (488, 561, 633, 405 nm) and three lasers (488, 633, 405 nm), respectively, and can be used for a broad range of multicolor measurements.<\/p>\n<p>&nbsp;<\/p>\n<h4>Contact<\/h4>\n<p>Dr. rer. nat.
